As a result of the evolution and inheritance of Y chromosomes the male lineage provides a means of biologically tracing and linking successive family branches. From analysis of DNA results from only six Le Couteur descendants so far, a framework of the three major branches has been established which is consistent with the known genealogy.

Other sub-branches can therefore now be generally confirmed by comparing an individual's Y-STR results to those that are already known. Candidates require a continuous male lineage to an ancestral Le Couteur on this tree and will generally have retained the surname Le Couteur. Tests from 3 different laboratories over 12 years have been found to be reliable and consistent.

Online results for three individuals are currently on the FTDNA database from their ‘37-marker panel’ Y-STR test. This detail, with that of 3 others, has confirmed the family branching, but ‘23-marker panel’ data are adequate to unambiguously resolve one’s biological Le Couteur identity.
